Like most of you I have many but I think my favorite is rom "One Eyed Jacks" when Karl Malden as dad Longworth replies to Brando's Kid Rio, "Oh sure, kid, you'll get a fair trial. Then I'm gonna hang you."
Follow Ups:
Uncle Benny on one of the Lethal Weapon movies.
Yoda: You must unlearn, what you have learned.
Darth Vader: Obi-Wan never told you what happened to your father.
Luke: He told me enough! He told me you killed him!
Darth Vader: No, Luke. I am your father.
Luke: "No thats not true, thats impossible!"
Darth Vader: "Search your feelings you know it to be true"
Luke: "Noooo, Noooo"
Princess Leia: I love you.
Han Solo: I know.
Yoda: ...for my ally is the Force. And a powerful ally it is.
Leia: I thought you decided to stay.
Han: Well, the bounty hunter we ran into in Ord Mantell changed my mind.
Leia: Han, we need you!
Han: We need?
Leia: Yes.
Han: Well, what about you need?
Leia: I need? I don't know what you're talking about.
Han: You probably don't.
Leia: And what precisely am I supposed to know?
Han: Come on! You want me to stay because of the way you feel about me.
Leia: Yes! You're a great help to us. You're a natural leader.
Han: No! That's not it. Come on. A-ha!
Leia: You're imagining things.
Han: Am I? Then why are you following me? Afraid I was gonna leave without giving a good-bye kiss?
Leia: I'd just as soon kiss a Wookiee!
Han: I can arrange that. You could use a good kiss!
I'm going to list all of Caddyshack then! or Fletch :)
From Raising Arizona:"No mam. We're not escapees, we released ourselves on our own recognizance".
"Hurry up. I'm in Dutch with the wife."
Big Lebowski,
"Man, that rug really tied the room together."
...do you work out? "Sort of...." Bud Bundy's response while giving a backrub to his neighbor's niece. Doesn't really count as a movie, but there are so friggin many good movie lines to list, some movies are just filled with them(i.e. nearly everything from Rodney and Knight in Caddyshack)
From "Silverado"
Sheriff Cobb: We're gonna give you a fair trial, followed by a first class hanging. From "Outlaw Josey Wales"
Josey Wales: When I get to likin' someone, they ain't around long.
Lone Watie: I notice when you get to DISlikin' someone they ain't around for long neither. From "Full Metal Jacket"
Gunnery Sergeant Hartman: How tall are you, private?
Private Cowboy: Sir, five-foot-nine, sir.
Gunnery Sergeant Hartman: Five-foot-nine, I didn't know they stacked shit that high.Gunnery Sergeant Hartman: Were you born a fat, slimy, scumbag puke pieca' shit Private Pyle, or did you have to work on it?
Gunnery Sergeant Hartman: Pyle, you climb obstacles like old people fuck.
"One Eyed Jacks" has several great lines.1. Pancho: "Rio, you could spend the rest of your life looking for
this man.
Rio: "Let's get goin'".2. Rio: "Don't be doin' her like that".
3. Rio: "Get up, you big tub a guts!"
Just crushing.

Grins
Such as:"Beulah, peel me a grape."
"Is that a banana in your pocket or are you just glad to see me?"
"Goodness had nothing to do with it."Need I go on?
Is a frog's ass water tight.
But I DO say no more than ten to twenty million killed, tops. Uh, depending on the breaks."
Most people don't understand the significance of that line - it explains every deus ex machina moment in the film (how they survive every explosion, every car crash, the high altitude demise of the nazis, etc.)

John Wayne in, I think, The Yellow Ribbon.
The wagon train is surrounded by savages.
Some young lieutenant volunteers to cross the lines and bring help.
Wayne watches him get caught in a cross-fire of about 500 arrows and, with his arms around the lieutenant's wife's (widow, now) shoulders he says, looking into the distance, "He would've wanted it that way."
In Commando (I hope I have the movie correct) Arnie is holding a guy over a cliff to get information out of him and tells him something like "if you tell me everything I'll let you go". After the guy tells him what he wants to know Arnie drops him off the cliff. A couple minutes later on the female character (I think) asks what he did with the guy. "I let him go".
I don't know if it's my favorite, but do remember it even many years later after one (somewhat painful) viewing.
Earlier in the film Aaaahnolld says to the same charachter " I like you Sully, that's why I'm going to kill you last." Another good one from that film is in the plane just before take-off. He breaks the bad guy's neck in the seat next to him, pulls a blanket up to his chin and says-"Stewardess, don't disturb my friend;he's dead tired."
I remember the first one now. Hmm, maybe I'll have to rent this one again one day.
[After throwing him off the building]Stone: Where is Nitti?
Ness: He's in the car.
