As for video, there is no "best" as long as you use them as they were intended. There recently was a "shootout" in one of the HT magazines, but I don't think they really picked a winner. If you get Video Essentials or Avia or both, you can't go wrong. There are some audio test signals on both of these discs, but the Stereophile test discs and a few others offer more audio-specific tracks. I have a Denon PCM calibration disc that I use every once in a while. As far as the "best" place to get it, do you mean the cheapest/least expensive? Try express.com, dvdplanet.com or search the online DVD sites.
