No. The 700 year-old knight guarding the grail says that it could not pass beyond the seal (in the Petra building) and "that is the price of immortality". Also, the knight's two brothers left Petra and died "of extreme old age".
So I assume in the movie's logic that Jones would have to keep drinking from the grail at regular intervals to remain "immortal". He only drank from it once. Perhaps he got a few extra months?
